I took out the comma before 'and'.
In answer to your question, the concept of freedom of religion, as it relates to the U.S. constitution, means that people may practice their religion free of government interference. It also means that the people may NOT, under any circumstances, try to establish an official religion, backed by the government. The only way that could happen would be to ammend the constitution, which is not likely.
As much as possible, the government must remain neutral in matters of religion. In reality, this doesn't happen all the time, but that it the tendency, at least.
On point that needs to be made - you may practice whatever religion you like in the U.S.; however, you don't have absolute freedom if the practice violates major laws. To give you an absurd example, you couldn't practice human sacrifice because that violates our laws agains murder.
