Can I use 'more' instead of 'again' here?

He used to be a dean of high school, but he isn't again now!

    "He used to be the dean of a high school, but not any longer"

    not really, i would rather say it this way:

    He used to be a dean of (our) high school, but he no longer is now.

    No, you cannot use it in this context. You want to say:
    "He used to be a dean of high school, but he isn't any more"
    "He used to be a dean of high school, but no any longer"

    :)

    "He used to be a dean of high school, but he isn't again now!"

    Instead of again you can use any more and remove the now. Your sentence would say, "He used to be a dean of high school, but he isn't any more."
