Which one is the right way to ask about someone's family? Are there other ways to ask it?

基本的にどちらでもOKです。
但し、目上の人と話すときには、「ご家族は何人いらっしゃいますか?」といった方がいいです。
My answer is both OK.
BUT, if you talk to your teachers, bosses, elders, ....etc, you need to talk in a more polite way. Ex: "Go-kazoku ha nannin irasshai masu ka?"
