    It should be "is". Who told you to use "are"?

    Unless, of course, the word was meant to be "relationships".

    I believe that this is a so-called "typo." That is, a typographical error.

    The writer wanted to write "What IS the relationship between the two speakers?"

    P.S. Perhaps the writer (a non-native speaker?) got confused by the phrase "two speakers."
