No difference in the meaning, but the gender of the subject that comes along with the word должен / должна / должно / должны is important.
Иван должен работать. Я должен идти. (m)
Анна должна работать. Я должна идти. (f)
Мы должны работать. Дети должны учиться. (pl)
Кафе должно быть здесь, недалеко. Это должно быть трудно - вставать каждый день в 6 часов. (n)

The problem is where is stress in должно. If it is должнО, Tati is absolutely right, it is a neutral short form of an adjective "дОлжный".
But there is also an adverb дОлжно. (Dative case)+дОлжно+infinitive. It is synonym of нужно, but дОлжно is literary and elevated style word. It is often use in theological books and by priests. For other people this word is old-stylished word.

Теперь у меня есть Библия и я знаю, как мне дОлжно поступать.
