how about "I know about 4000 words of English"?

If you substitute 'an English vocabulary', for 'a English vocabulary', then 1 is okay, as is 2. You could also say, "I know 4,000 English words", or, "I have a vocabulary of 4000 English words"', or "I have an English vocabulary of 4000 words".
