Let’s say you are at an airport and there’s a line for people to be check in. Let’s also suppose you are with a friend but you two bought the tickets. Then, while the attendant is checking you in, you say, while pointing to your friend.

“Sir, after your are done/finished with me, could you do him?”

Does it sound odd to use “do him” here? If so, what could I say? “Get to him”?

I agree with Lizzy's comment. Another way to express it would be, "Sir, after you are finished with me, could you please take care of him next?" It's very polite and won't cause any confusion.

'Do him' is ok – we will definitely understand what you mean. However, it is very informal and does sound odd (partly because 'do him' means 'sleep with him' in certain contexts). Native speakers only say 'do him, do me next, etc.' when we are lazy or in a rush, and can't think of a better way to express the idea.

''Could you please help my friend, after me?  Its just that you give great service''.  that last part is something I notice people do , to sweeten the proposal

To continue the airport example, sometimes people get frustrated if they have to wait in a long line to check in. If an employee sees that people are becoming frustrated, they might say, 'Thank you all for your patience. We will get to you as soon as possible.' The people in the line might say, "I hope they get to us in time, otherwise we'll miss our flight!" So, it doesn't make much sense to use 'get to him' in the context with your friend because you have already gotten to the front of the line and are being helped by the attendant, so helping your friend will be the very next thing the attendant will do.
