I'm not a specialist. But, I´m brazilian, i've been to Portugal and I read books edited in both countries. I think the main difference is the accent and the formality. Portuguese spoken and written in Brazil is usually more informal. There are a few different words too.

There's no only one difference, but many diferences and some of them very deep and most in the spoken language. For example, when speaking, brazilian portuguese pronounces the vowels strongly than the consonants. In portugal it's exactly contrary. That's why, sometimes, people from Brazil cannot undestand people from Portugal. Writing and reading it's almost the same in both language variants.
