I heard a line from TV "The Good Place" : Oh, Ariana. We're really in it now.
What does "really in it now" mean ? Does it mean we're in trouble?

That is a very good show.

Yes--it means they are in too much trouble to try to pretend they are not in trouble. That show is great, but there is a lot of casual and colloquial speech in it!
