What if I were to say "take a breathe" instead of "take a breath"? Would it still make sense? Thanks

BREATHE is a verb, so you can't take it. BREATH is a noun.

No, but you could say "take a breather" if you are referring to stopping what you are doing for a short time to have a rest.
