You can never say 'most of people'.
You can only say 'most of' before a pronoun.

If you are talking about a large or general group then you need to use( most people ):
Most people know how to write a letter.
But, if you are talking about a smaller or more specific group then you need to use ( most of people )
Most of people in my neighborhood are well educated.

Or as in Fahad's example sentence, you would have to say "Most of the people in my neighborhood are well-educated."

Yes, I agree with Jmat, you can never say "most of people." Just "most people" OR "most of them."
