Is it appropriate to say Happy Mother's Day to a man in USA?
Some of my colleagues say there is nothing wrong with that. In cases when a man totally takes care of a child/children (i.e., there is no mother), then I assume it is appropriate.
But what if a man doesn't have children? is it still ok?
In Europe there is Women's Day, March 8th. It is NOT appropriate to say Happy Women's day to a man; he will get upset.
What about Mother's day in the US? Will a man who doesn't have children get offended if one says Happy Mother's Day to him?

No, you would not say it to a man, and generally you would say it only to a mother.
It wouldn't be considered insulting, nobody would be upset, but it would be considered a funny mistake.
Fathers get their chance on Father's Day, June 21, 2015 in the United States.
As to who counts as a mother, I would guess that if a child calls somebody "mother," then they are a mother and are entitled to recognition on Mother's Day.

For the most part Yes, usually you don't say Happy mother's day to a man but I'm sure there have been instances that it has happened. All in all, if I were you, I wouldn't
