"What is in the box" or "What's in the box" will do just fine.

I think a good general rule is:

What is + [noun/adj phrase]
What's there in the box?
What's the matter?
What's wrong?
I don't see what's special about this sentence
---
What is there + [verb]
What's there to do in Spain?
What's there to complain about?
What's there to eat in this restaurant?
