No. Nobody does.
It's a set phrase that only exists in the negative.
Someone who 'doesn't suffer fools gladly' has no time for stupid people. Think of it as a fixed expression.

Usually (always) used in the negative: He/she doesn't suffer fools gladly = He/she has no patience with fools. ( = does not like the company of fools)
The question "do you suffer fools gladly?" doesn't exist.
