Hi! Only the first one is correct. Because it is the question to the subject.

They are both grammatically correct, but the first one is what you should use most of the time. In the second one, the addition of DOES makes it more emphatic. Example, "My father doesn't know the answer, my mother, brothers and sisters don't know the answer. Who DOES know the answer?"
