This is my colleague & This is a colleague... This is my colleague. & This is a colleague of mine. What is the difference between sentences?

There is no practical difference, except the second one suggests you have more colleagues than just one. 'My colleague' has the potential to mean 'this is my only colleague' but it is not implied. Both are good and sound natural for introducing your colleague.
