Asking the customer about the laundry.
As a hotel front office staff, how do I ask a customer if he has clothes in the laundry service?
Does "Do u have clothes in the laundry service?" sound natural?

It sounds fine.
The only mistake is the spelling of the word 'you'. Remember that the only one-letter words in the English language are 'I' and 'a' - there is no such word as 'u'. Your question should be written "Do you have clothes in the laundry service?'
It might also sound more natural to add the word 'any' before 'clothes'.

Hi Graham. Do you have clothes at the laundry service. Remember preposition of place. Again it's confusing even for me. I take my clothes to the laundry. My clothes are in the washing machine. My clothes are at the laundry/laundromat.
