Can we use this sentence when we want to fire at someone? " Let me cancel him out"

You probably could use it but I've never seen it used to mean fire at and kill someone. It would be better and is more common to say 'Let me take him out.'

Yes, but he might first "cancel" you out whilst you are chatting, since a bullet can fly faster than five words.
