If someone use " in my department" to describe his job
If someone use " in my department" to describe his job, does that mean that he is the supervisor of this department?

Claire is right. Just as when we were studying in a school, we used to say "In my school ....." which of course, only meant that we were a student of the school rather than the head master or the owner :)

No. It could be that they just work in the department.
