When can we say this sentence? "She is too happy to speak." Does it mean "she is very willing to speak" or "she can't speak because she is so excited"?

The second one is the right meaning. I think the other phrase you are thinking of, which means, "she is very willing to speak," is "she is more than happy to speak."
