I could probably give a better answer if I knew the content in which the sentence is being asked but it is most likely going to be "are you in line". For example: Are you in line for the cash register? You would probably use "are you in this line" before you use "the line" Hope that helped :)

We use either. It's not a style choice - it depends on the context.
