Is it a car of yours or is it the car of yours? Which one is correct: Is it a car of yours? or Is it the car of yours? For example, one person talk with another one and he points his finger at the car.

No one would ask, "Is it the car of yours?" That is not a natural expression. To ask "is it a car of yours?" implies the person is believed to own more than one car. That's certainly possible, but it would still not be the most natural way of speaking. Do as Dave suggests.
