In this context there is no real difference. The use of "mailed," might imply that the speaker is handing somebody a letter than has already been put in an addressed envelope whereas "that you should send," might imply that the speaker is handing somebody a piece of paper from the printer. But the overall meaning is exactly the same - put this letter in the post ASAP.
