Do you know the time of your next flight? Do you know the time of your flight? (Both “departure” and “next” would be the default assumption)

Yes that's correct. You could also say... Do you already know when your next flight will depart? Do you know the departure time of your next flight? Did you find out when your next flight will depart? They all mean the same thing and they are all equally correct.
