Negative sentence
Hi, there
I know you can say “Which countries do American citizens require a visa for?”... But what if i want to make this sentence negative? Should I say “Which countries do Americans not require a visa for?”

As a native speaker I would have to say your sentence sounds just right. :)
Which questions do people not required answered?
Which pastimes do not require a lot of dexterity?
Which sports don't you play?
“Which countries don't Americans require to have a visa?” <-- not nearly as neat.

As a native speaker (not a grammarian) I think your second sentence is correct, and it would probably be understood. However, it sounds awkward. As with many such problems, I would sidestep it by rewriting the sentence in a completely different way. For example:
"Which countries can Americans visit without a visa?"
