Basically when I phone a person Its not the actual person whom Im looking for its his/her assistant or its a person who is replaced for a moment.

I start like this:
Me:Bonjour. C'est X, "Puis-je parler à Mme.Y" / "Est-ce Mme.Y est là"?
On phone:Bonjour. Elle est absente

I'm looking for more options to replace the texts in inverted commas.

Je suis Monsieur/Madame X
Je souhaiterais/voudrais parler à Madame Y, est-elle là/est-elle au bureau?

"Puis-je parler à Mme.Y ?" (good) 
"Est-ce QUE Mme.Y est là"? (the word "que" was missing)
"Je m'appelle ... J'aimerais parler à..."
"Pardon de vous déranger, est-ce que je peux parler à..."
"Pourriez-vous me dire si ... est là ? Est-ce que je peux lui parler?"
