No existen reglas exactas para el uso de los verbos "ser" o "estar".
"Ser" es un verbo que define el sujeto es decir, señala una característica que puede ser permanente o accidental, ya que eso no es de mayor importancia. "ser" identifica el sujeto y el atributo de una oración.
Se puede utilizar en lugar del verbo "existir".
El verbo "estar" es un verbo de situación, resultado y estado. Con este verbo expresamos en Español la situación en el espacio y tiempo, circunstancias, estados, intenciones, momentos de una acción etc.
El verbo "estar" es un verbo de "estado" que señala como se encuentra en un momento determinado o siempre, el sujeto.

WHEN TO USE ESTAR:
For locations:
Ex:
I am in the city - Estoy en la ciudad
He is at the park - El esta en el parque
For moods that aren't permanent:
Ex:
I am sick - Estoy enferma
*You're not sick all the time, that's why you use "estar".
She is drunk - Esta borracha
*Here you're saying at the moment she's drunk, but she's not drunk all the time.
Esta guapo - He is handsome
*Here you're saying that right now he looks handsome, and not necessarily handsome in other days. Equivalent to--> That guy looks good today for some reason, what did he do to improve himself?
WHEN TO USE SER:
For describing something that is permanent:
Ex:
She is drunk - Es borracha
*Here you're saying that this girl is a always wasted and is definitely labeled a drunkard.
He is handsome - Es guapo
*In this case you're saying that this guy is handsome all the time and not just on certain occasions. Equivalent to-->That guy is hot!
We are teachers - Nosotros somos profesores
*With occupations you need to use "ser"
OTHER USES OF ESTAR:
Verb in the present tense can be conjugated in two ways:
I sing OR I am singing
The same goes for Spanish:
Yo canto OR Estoy cantando
Estas corriendo - You are running
Esta hablando - He/She is speaking
Estamos comiendo - We are eating
Estais durmiendo - You (Plural) are sleeping
Estan bailando - They are dancing

Estar se usa cuando hablas sobre un lugar posición o estado de animo
por ejemplo
"alguien debería estar leyendo esto"
y ser mm no se me ocurre como definirlo asi que pondré ejemplos =)
"esto debe ser un error"
"tú debes ser quién estoy esperando"
"debemos ser optimistas ante todo"

you use "ser" for things that currently ARE and have been that way for a long time or things that will be that way for a long time.
and "estar" for things that currently ARE but have just turned that way recently and aren't expected to remain like that for a longtime.
I am alive = Estoy vivo ( I will die for sure~ )
