''assume/on the assumption...' ''I assumed that he was much older than my sister.'' ''I was on the assumption that he was much older than my sister.'' Do they have the same meaning? Can I use them interchangeably?

We don't say "be on the assumption." You do something "on the assumption" that something is probably true. Example: I acted on the assumption that he was much older than my sister. In this case, I took an action based on my belief that he was much older than my sister. "I assumed he was much older than my sister" simply expresses what the speaker believed was true.
