What does it mean?

I want to know what does people mean when they say this. Like Chuck said to Blair in Gossip Girl, and i've heard this in Friends.

It's a joke! Actually the only word with these two pronunciations is "tomato".
1. BrE = to mah to
2. AmE = to may to
"Potato" only has one pronunciation: "po tay to". People use this word as a kind of joke.
It is also a kind of joke on how uneducated some Americans are. They may actually believe the English pronounce it "potahto".
