"You and I are friends." is correct. You'll hear native speaks say "You and me" or even "Me and you". These are both incorrect. "I" is a subject pronoun. "Me" is a direct,indirect, or disjunctive pronoun. It cannot act as a subject unless you are talking thereabout as a word (like what I did in the last sentence.)

I think both are right, but the first one is more correct becasue you use the pronoun 'I' before the verb ''to be'' It's like you say ''you are my friend'' ''I am your friend'' = we are both friends = you and I are friends

You and I are friends. You and me are friends. Both these phrases are correct. The use of "me" in this context is too complicated to explain. But it is correct.
