What's the difference between "Get off me" and "Get off of me" when I tell someone to let go of me?

No difference both are fine, but "Get off me" is the more common expression

There is no difference its just that "get off me" is mostly used by the British English-speaking and "get off of me" is mostly likely to be used by American English-speaking. Both terms are used and they mean the same thing.
