Often, the way the two phrases are used means the same thing. There is a subtle difference. "Glad to speak to you" implies that one person is speaking and the other listening, with little or no conversation. "Glad to speak with you" implies a conversation, with all people involved speaking. As I said, in actual everyday usage, the two are used interchangeably.
