They are two different questions for different circumstances. "Do you wear glasses?" is a general question that implies "all the time" or "in general". "Are you wearing glasses?" is present and is asking if you are wearing glasses right now and not whether it is something you always do.

They both are correct, depending upon the meaning of the question. One may "need to wear glasses" yet not "be wearing glasses at the moment". "Wearing" is in the past, present, or future progressive tense.
