Do you differentiate between 层 and 楼?

For example, if I wanted to say, "this building is 20 stories tall = this is a 20-story building", should I say 这件建筑一共有20层 or 这件建筑一共有20楼 or are both correct?

Also, if I wanted to tell my neighbor that I live on the 6th floor, should I say 我住6楼 or 我住6层 or are both correct?

All your sentence are correct. However, 层 is a more accurate word when you refer to floors in a building.
