"Stairway to Heaven" is a very famous song considered to be a classic. The joke means that this "break-up line," the excuse used to end the relationship, is just as classic as that song and can't be overused. As far as the "of"... It is used the same way as "he is the leader of the group" or "he is the genius of the family."
