In American English, you would say, "Are you in line?" if you want to make sure you are not breaking in front of someone. Do not say, "in the line".

In American English, one would ask: "Are you on line?" The word "queue" is used more in British English. In the scenario of waiting at an ATM, one might ask: "Are you waiting to use the ATM?"
