You can use both. But with "What is your Job?" I would only use it in more friendly and colloquial situations e.g. talking to someone you just met at a party i would use the du form "Was machst du?". In setting were i would use the polite form already i would may be rather say something like "Was machen Sie beruflich?" Beruf = Job I hope that helps :-)
