Kill is a general word... kill off has particular meanings:
In literature, it is when the author decides to kill one of the characters in the story and continue the narrative without it...
You also use kill off to say killing a big group of something

Yes, there is! "Kill off" refers to killing an entire group. "The dinosaurs were killed of by an asteroid."
