Hi... The meaning is the same one is informal (nope) and the other one is formal (no) ...
In a writing for example don't use "nope" use it only to a friend in a chatroom

nope (informal) = no
It is the opposite of 'yep' (= yes).

the difference in between no and nope is that, no is in pure British english and nope is in American and nope also adds to the latest trends.
