Usually we say:
Who is the best singer?
Who is the best athlete?
Who is the best teacher/student?
In the 3 examples above, you need a noun after 'best' = the best what.
"Who's best" is short form of "who's the best" and it is usually used when talking.
You would always be correct to use "Who's the best?"
