In college settings, each class you take is on a subject. The area or subject you specialize in is your major. For example, if you want a BS in mathematics, you will major in mathematics. Some classes you will have to take in this major are Calculus and Differential Equations. The names of those classes indicate the subjects. Does that make the distinction clear? Are were you speaking of a scholastic context?
