Is this sentence correct ? He treats people most politely in his class. Why is there not a the in this sentence before the word most. Thanks for your help.

The sentence is correct. The word "most" in this case is used as an adverb of degree, like "very" and not as a superlative, as in "the most polite person". It is a rare but possible use of the word and is usually formal.
