is there any substance in their claim? is there any substance IN their claim? or is there any substance TO their claim? Which one of these two sentences is correct?

"to their claim" is correct You'd hear "in" more like this... Is there any substance in what they're saying...

Either one is correct. You would hear 'substance TO' more often, but both would be understood.
