I am off from Thursday till Friday.
I am off on Thursday and Friday.
Which one is correct or more native?Can I use off as preposition and say " I am off job " to mean that I am currently out of the office .
The second one is better, however you can make it even more informal and natural: "I am off Thursday and Friday.'

1st sentence: it would be better to say "I'm off Thursday thru Saturday".
("I'm" is putting together "i" and "am", which is how native speakers actually talk. No one would say "I am" unless they were being very formal.)

The phrase you want is "off work". No-one uses "off job" because it's broken English.

Both are acceptable, but you can't say "off job". At least "taking off from my job" sounds better, informally.
