I agree with Colin. One and three do not sound natural because of the repetition of "lead(s)". You could have said: "... a clue that leads..." or "... clues that lead...". or "... evidence that leads...".

No. 2. Next would be No. 4, but the plural "leads" suggests that only one lead is not enough to be hoped for, and in this context I think it certainly is. Any lead here is a great thing.

A ‘lead’ is something that you need to begin the process of finding more significant evidence. You don’t look for leads, you have them and then follow up on them. A lead is evidence which might lead to further evidence. We have a dead body and no leads. (We can’t make progress) The only lead we have is an eyewitness who saw the deceased leaving his apartment with four men. (We can try to find out who this group of men was) I want you to follow up on all of our leads, no matter how unpromising they might seem. You can use ‘lead’ as a verb. For example: The evidence led us to you. The evidence led me to believe that this was a crime of passion.
