What type of people would you use the first sentence with? A university professor? Boss? As an employee to customers? Thanks!

To someone who you would normally speak to formally, someone who entails a great deal of respect, you would use 您.
In an informal situation, you would use 你. Both terms, however, retain the same meaning, which is "you."

您(You) is referring to someone you respect. Yep university professor, boss, customer, teacher can all use this.

您好吗? (this is a formal so you can use to old people that you and someone's a career like teacher, professor, engineer and boss ).
你好吗? (that is informal so you can use to young people and friends)

Okay, we always use "您" to show our respect to someone such as professors, teachers, of course your boss, and maybe the old. But in fact, now in China, more and more people pay no attention to the character :)
