In English 'hon wo yonde' is the imperative form. You are ordering someone to read (i.e. Read the book). "yomimasu" and "yomu" are actions that you do. "yomimasu" is a polite way to say I read the book (repeated actions) or I will read the book. "Yomu" is plain form and can be used with friends and family or people you know really well.
Hope that helps. Enjoy your study!
