What preposition should we use here. this dress looks great on/of/over you.

'on' is the best preposition in this sentence.

Yes, "on" is correct. "This dress looks great on you" is perfect.
However, from the point of view of human relations, it may be better to say something that has a slightly different meaning: "You look great in this dress."
