Which sentence is correct?
1. Spanish is spoken primarily in Latin America and Spain, but also in other parts of the world with Hispanic POPULATION.

Only the second one is correct. In this context, the word "population" is a countable, singular noun, so it needs to either have an article - as in 'a Hispanic population' - or be in the plural without an article. Here, the plural makes sense because you are talking about several populations in different countries.
