Are both variants possible or only the first one? - "the friendliest" and "the most friendly"

It is possible to use 'more' and 'most' with two-syllable adjectives ending in 'ly'. Your choice will depend on what fits best into the sentence. Here's an example where the 'most' version is preferable:
"She's the most friendly and likeable girl in the class."
'Most' is a good choice here, because it works with both adjectives.

Both are correct in terms of grammar. You can use both. Personally I use both, unless it comes at the end of a sentence. At the end of a sentence I tend to use "friendliest" (this is just a personal preference) For example:
She is the friendliest person you will ever meet.
She is the most friendly person you will ever meet.
My pet cat is the friendliest.
